Seb Larsson struck the only goal of the game to sink Ryan Giggs' side and help Gus Poyet's men stay one step ahead of the drop zone.
Despite dominating the ball for the first 30 minutes, United fell victim to Larsson who steered home after being played in by Connor Wickham.
Sunderland continued to surrender possession but United were unable to create any sort of chances as the Black Cats sat deep and remained tight.
United felt they should have been awarded a penalty after half-time for handball, but Nemanja Vidic's protests were turned down by ref Howard Webb.
Sunderland continued to look dangerous on the break, and Emanuele Giaccherini's powerful effort looked to have beaten David De Gea before striking the post and bouncing out of play.
Fabio Borini then struck the woodwork for the Black Cats, leaving United rattled.
Giggs' men pressed hard but couldn't find a way through, leaving Sunderland to claim all three points.
The win sees Sunderland move three points clear of the drop zone, and with two games remaining they will be confident of retaining their Premiership status.
United remain in 7th, six points off Tottenham, but will hoping to make up the ground in their final two games.
